02 // But the Sky is Clear

As a faithful Californian, no vacation during Winter would be complete without a trip to somewhere cold. You know, to escape the warmth and sun of Cali. I mean, perhaps I could go somewhere warmer and sunnier, with higher Uggs-to-girls ratio, with more In-N-Out burgers, and with happier cows that produce better milk. But then you'd just be in a different part of California. So Boston.

Boston is frigid cold now. If you actively check web-aggregate sites like Digg.com or reddit.com, you'll have seen pictures of Britain painted in white by snow, and the news on the ground is as bleak as the photo. Winter explains some of it, yeah, but there's another chilling factor (see what I did there?). The rest comes from something called the Arctic Oscillation (AO). The unusual chill is due to the AO entering a negative phase, whereas it had been mostly in the positive phase in recent history. Typically, in the positive phase, we'd see lower pressures in the north, resulting in the cold being kept out of most of the mid-northern US by a buffer of high pressure. But since now the higher pressure is north of us, we get a blast of chilly air from the Arctic. This alternation of phases is normal according to meteorologists, but it seems novel to us since the most recent phase shift prior to this year's event was in the 1970's. The new effects will be noticed mostly by those living in Midwest United States... and the Arctic (which is consequently warmer than usual). I wonder how the polar bears will fare this year with a limited amount of ice formation...

Let's not think about the diminishing polar bear population and instead discuss something more fun, like biochemistry. In a class I took called "20.320, Analysis of Biomolecular Systems" (no really, I'm serious, we're talking about biochemistry), I had a project in which I had to model a regulatory pathway of my choosing that would ultimately increase or decrease under certain conditions the expression of a protein - in this hypothetical case it was GFP. I chose, rather foolishly I suppose, what I thought at first to be a simple system involving microRNA (miRNA). miRNA is a small strand of RNA (around 21-24 nucleotides long) that thus far has been found to "silence" gene expression by binding onto mRNA, thus disrupting translation. In summary, I hypothesized that miRNA that targeted the gene expressing GFP would be increased by a transcription factor (the transcription factor binding step was not important for the model), but would be regulated downstream during translation by the disruption of miRNA binding to target mRNA, consequently increasing GFP expression, which makes sense - disrupt the disruptor and you get positive activity. It turns out there have been several studies on endogenous regulators of miRNA silencing (called RNA interference in general), most of which involve either the dicing of miRNA itself or the simple removal of miRNA from the cytosol (by specified ribosomes). At this point I saw it would be more efficient if there was some other mechanism that would link miRNA production to GFP DNA production so that so much miRNA and DNA didn't have to be wasted, but I couldn't find any relevant literature. Having no other explanation at the time, I was begrudgingly satisfied and went on with the project, which ended up being a pain versus a simpler model I could've chosen, which was a model of transcription factors similar to the lac operon (but everyone else was doing this, and I didn't want to be everyone else! House. Road House.). The pain was that at the same time as GFP was being produced, miRNA was also disrupting GFP production, which made the system a parametric one of dimension n-squared versus a linear system... and I had to solve the differential equations for all of that shit (about 18 different equations with 18 unknowns in all)!

Anyways, why I'm talking about this now is that I read in the most recent Cell issue about a study that could have majorly simplified my model and also my life. This: Transcriptional Control of Gene Expression by MicroRNAs. To summarize, Basel Khraiwesh et al found that miRNA use epigenetic control by methylating DNA transcription sites (making them inactive) to regulate gene expression at high concentration. They also found that at low concentration, the standard RNA/translational control applies, which says this mechanism may be dependent on the miRNA-to-target ratio. The study was done in moss, but the hope is that it translates to mammals. I'm impressed because this also implies that instead of seeing miRNA as a second-son of gene regulation relative to transcription factors, it's just as important, if not more so. Now if only they'd published this a year earlier...

I braved the cold to seek liquid warmth in a bar crawl with a few friends in Boston. Before the expedition began, we determined the best way to know if a place is good is by the quality of their french fries, a true American staple. We found that none of the places we went to had fries. I greatly underestimated how much the East Coast sucked.

After three American religious representatives spoke at a seminar in Uganda, the government passed a legislation that punishes homosexuality to the extent of, in some cases, execution. It turns out that two of the three implicated lecturers were from the Sacramento area. To his credit, one, Don Schmierer, says he is greatly repentant. A law that puts human life at risk without imminent threat with proof of causality is a bad one. A law that imposes a way of life, no matter what the cause, is a bad one as it opens the gate for other "social laws" that would soon impede on the freedoms once thought natural to all citizens (the classic "slippery slope").

He's produced some nice kpop remixes, usually infusing them with an electronica feel or with an American pop song. Aliks (aka Tae, "Bug"), is a Seoul born jockey with a knack for big beats. His songs take the underlying beat of the original song and adds bells and whistles to give them a "RedOne" (clearly one of his major influences) makeover.

Stopping by Barnes and Nobles (on the way to Best Buy) I got a book I've heard about and meant to check out. Of course the title of the book would be something like: How to Survive a Robot Uprising. Asking the B&N staff where to find it was awesome since the clerk didn't even blink. In fact she was a fan and we had a conversation about how to survive a zombie apocalypse. On that note, I need to procure some MREs and a police baton because we all know that police batons do the most damage to a Tank in Left 4 Dead 2, up there with frying pan. The book delivers. If you need any pointers on how to notice if your household robot is going rebellious, ask me and we can meet within a Faraday cage to speak details.

Digging through some old tracks, I found the original samples taken for Beyonce's "Crazy in Love" and a couple of Jay-Z's songs, most notably "99 Problems" and "December 4th". The former takes cues from The Chi-Lites' (hehe, no relation) "Are You My Woman?". It's pretty obvious where they sampled :) Hip Hop

Besides gearing up for archery (the backyard has squirrels and birds aplenty. JK!) and taiko, I've been getting into selling stuff on eBay. And so has my dad. I'm going to make it a friendly competition and say so far that I'm winning with a whopping $20.05 profit, although he recently put up my Buffet clarinet for ~$1700... that's going to take some work to catch up on if it sells.

Celebrating the New Year in Sacramento for the first time and having friends meet up in San Fran was a blast! I hope you all have a Happy New Year and pick reasonable resolutions.

Weeka-pedia #001 (Japan: hip hop and ukiyo-e)

01 // In

These music videos highlight an interesting cell-shading-type animation style that is similar to, if not modeled after, the Japanese ukiyo-e style. This makes for a visually intoxicating (fun) and trippy experience. All of these videos are Japanese groups, and I'll explain as you move through. Go ahead and ~~do whatever it is you do to~~ prep for this, it won't be enough!

HiFana - Wamono
HiFana is a beat-mixing duo that features percussion instruments (including a full drum set), MPC, and turntables. They pioneered with Pioneer (the company, heh) the DVJ, or DVD/CD turntable deck, and are associated with a break-heavy, hip-hop style. The title of this song, Wamono, can be translated to mean "anything traditionally Japanese", which makes it the perfect starting place (totally coincidence I linked it in this order, but hellawesome!) in scoping out the ukiyo-e style.

nobodyknows+ - Kokoro Odoru
nobodyknows+ (not capitalized on purpose), is a Japanese band whose style is characterized as a mixture of hip hop, pop, and rock. Their music carries a fast, bouncy beat with some synth and guitar sounds and a layer of rap to balance everything out. Their songs are consistently exposed to Japanese popular culture in the form of anime TV series, videogames and movies, most notably as part of the release of "Kung Fu Hustle" in Japan. This video's more tame/conventional. Kokoro Odoru is about the joys of music and dancing (hence all the "Enjoy!").

ASIAN KUNG-FU GENERATION - Atarashii sekai
ASIAN KUNG-FU GENERATION (in perfect contrast to the previous band, this one uses all caps) is a 4-member Japanese rock band that features a punk-influenced alternative rock style. They also feature a rhythmic lyrical style over a fast beat, but their rock-edge is harder with more guitar riffage and melodic vocals. Atarashii sekai means "new world"; the song is part of an album that revolves around a concept world. This video is a step up in trippiness from the previous, but is still no mermaid b00b.

02 //N'

I found a small records store near downtown and went digging. There were a lot of cool jazz LPs/EPs as a decent collection of rock/pop. They also had a table full of boxes with really cheap singles or albums they marked down to 3 for $1. $20 got me a Duke Ellington, Van Halen (not what you'd think is my cup of tea, but I like the heavy guitar, yo), a Destiny's Child single (really happy about this one), a Beatles compilation (I don't know what's on it, but I'm hoping it has "I'll Follow the Sun"), and a bunch of dollar records including two Japanese ones, one of which is a single by Ayumi Hamasaki!

Coincidence: while I was clearing out old stuff from the house, I discovered my dad's old stash of vinyl records, one of which was the other half of The Beatles compilation series (sweet). Besides a bunch of oldies (Neil Diamond, The Beatles, various pop stars of "the ancients"), there were a few that need to be mentioned from the collection.

In prepping for the return of my PS3, I've been playing a mad amount of CoD4 (the first one). I had to redo my entire profile, leveling up, unlocking guns, making classes, figuring out perks, etc. Add also that playing on the PC without auto-locking, non-normalized hardware specs, and a much quicker response time necessary to get the kills, it's a humbling experience. But I still got it. 7 hours played and I'm level 26 out of 55, beeotch!

Android making headway on iPhone - The release of the HTC Hero and Motorola Droid have been a relative success in Verizon's coup at AT&T and Apple's reign over 3G smartphones, and it looks like the war's only getting started. Among reports that people seek an Android phone (17%) as much as iPhone (20%), the balance of power for apps is also predicted to shift towards Android. Good job Google, breaking down tyrant-monopolies and becoming one, one product at a time!

Dark matter possibly detected - In the Cryogenic Dark Matter Search project, based within a Minnesota mine (to protect the detecting instruments from interfering space waves, i.e. cosmic rays) have a 1/4 chance that they have detected signs of dark matter (more specifically WIMPs). If this is confirmed, future devices measuring dark matter can help us find the source, which is expected to be the center of galaxies.

P.E.T.O.P. - Facebook group (might have to login to view): People for the Ethical Treatment of Pokemon. "A group of people dedicated to stopping the awful practice of pokemon battling, where people catch harmless, innocent fuzzy creatures and make them fight!" ... This one's a two-part piece. Masamix makes some sick mashups of Korean and Japanese pop artists often mixing them with well known American artists. Here are two features that mixes the basic top 51 in Korea into two songs; impressive transitions and continuity.

02 // Methods and Materials

I've basically bought as many magazines from the airport store as I found interesting since I figured I'd might as well get updated on the real world now that I'm out. These included:

I got through most of them because most of them were full of ads and not as much interesting content as the covers depicted, which is what I based the "interestingness factor" on for most of them (they get me everytime!). People and Popular Science contain the least impact/interesting per time spent, a unit of measure I'll call the "Chi Unit", or "X" for short.

The Chi Unit (X)

A measurement of interestingness per impact per time [interestingness / (impact x time)]. 1 X is equal to the feeling of walking to class in the morning during a rainy day whereas 1 MX (that's one megaChi) is something like snowboarding down a black diamond slope on top of fresh powder. You can extrapolate from these measures that reading my blog is somewhere around 1 GX (gigaChi).

03 // Results

Going through my room, purging it of all my old younger-days paraphernalia, replacing some, rearranging the whole place, helping Moms do chores that require so-called "manly muscles" as mine (sigh). She can't keep reminding me how much weight I've seemed to gain since last year.

Basically this is how it usually goes down:

Mom: "Before I was so worried; you looked like a boy. But now you look like a man!"

It's good to be home.

I also received a package yesterday, which surprised me since I wasn't expecting anything of mine to be in until next Tuesday. When I opened it, what do I find, but a copy of Modern Warfare 2! I've played it on a friend's console in Baker House, and I discovered a new talent and passion... apparently Modern Warfare 2 on console. There's only a few people I know who would send me something so awesome. But only one in that set with whom I'd gladly play some serious MW2. Zihao, if you know him (if you don't, then you should), sent me this sweet gift, and I can't thank him enough what it means let alone what it is. Thanks, Zihao!

My handle will be "Chi Unit" on PSN when I get my PS3 set up again.

04 // Conclusions

The New Super-Earth - temporarily called "GJ 1214b", the planet scientists discovered recently is the most-Earth-like planet to date, consisting of an atmosphere and a massive ocean. People are already submitting names to be considered for it. "Stephen Colbert" is #2, lol.

The idiot's guide to Google Wave - because we all don't know what the hell it's for.

Sexting is bad, m'kay? - a poll done by the Pew Internet & American Life Project showed as many as 15% teens "sext", or send text messages with sexual messages. Besides finding about a new word of which I've been missing the opportunity to utilize more regularly for several years, CNN and the Pew focus group found that "teens sext to people whom they are in a relationship, but often times (where the danger lies) [these messages] are forwarded to people outside the relationship, especially after a breakup." If it leaves a paper trail, don't do it, is what my Godfather used to say.

Tiger Woods scandal - I only keep up with celebrity news on the occasions when it involves people I like (i.e. the Rihanna scandal) or just simply blindside me (in essence it'd be or exceed a kiloChi). So Tiger Woods. Besides him crashing his SUV into a tree because he was on Vicadin and Ambien before he drove, women all over have been popping up claiming he had an affair with them. There are allegedly 7-8 women now implicated, according to People. This gives new meaning to the phrase, "Go get 'em, Tiger." The tragedy is with Elin, his wife, and their son and daughter, although most direct sources to Elin and Tiger say they're trying to get through the mess while preserving the family. It sounds like most of the credible cases with Tiger involve him contacting them frequently, although the ratio of romance to tryst of these meetings is still under scrutiny. A lot of these women surfacing are "party girls" and are probably just jumping onto the bandwagon for the money. Guys and girls screw each other equally in different ways.

03 // 'Sup:
The plane out of Boston was delayed due to a fuel line break and had to go under maintenance. The pilot said, “The ‘quick fix’ isn’t going to work.” It’s comforting to know they tried getting us promptly to our location despite a fuel line break.

PROTIP:

Flying a plane with a fuel line break is not advised. Carrying 3+ laptops into the security check is also not advised unless you want the nice lady behind you to say something like, "WTF, mang."

They transferred me onto the next flight to Chicago-O’hare, which meant I’d miss the connection flight to Sacto so they booked me on the first flight out of Chicago and checked me into a hotel there with $3 for breakfast (getting paid in full!). The hotel was decked out in Christmas gear and everyone was nice so I couldn’t get too annoyed. Also I won a Premier Line pass in the airport coming back in, although I already had Premier Exec… so I got playing cards instead. My dad, more awesomely, won 20% off his next travel itinerary.

I’m thinking of keeping a separate blog for more interesting subjects that I want to keep note of not only for myself (since I want to use my memory on more imminent issues), but for your input. I’ll post links to the updated blog on this guy when I put something new up.
