You know, to escape the warmth and sun of Cali. I mean, perhaps I could go somewhere warmer and sunnier, with higher Uggs-to-girls ratio, with more In-N-Out burgers, and with happier cows that produce better milk. But then you'd just be in a different part of California. So Boston.&lt;br /&gt;&lt;br /&gt;Boston is frigid cold now. If you actively check web-aggregate sites like Digg.com or reddit.com, you'll have seen &lt;a href="http://i.imgur.com/cYooH.jpg"&gt;pictures of Britain painted in white by snow&lt;/a&gt;, and the &lt;a href="http://www.independent.co.uk/news/uk/home-news/britain-facing-coldest-night-of-winter-1860407.html"&gt;news on the ground&lt;/a&gt; is as bleak as the photo. Winter explains some of it, yeah, but there's another chilling factor (see what I did there?). The rest comes from something called the Arctic Oscillation (AO). The unusual chill is due to the AO entering a negative phase, whereas it had been mostly in the positive phase in recent history. Typically, in the positive phase, we'd see lower pressures in the north, resulting in the cold being kept out of most of the mid-northern US by a buffer of high pressure. But since now the higher pressure is north of us, we get a blast of chilly air from the Arctic. This alternation of phases is normal according to meteorologists, but it seems novel to us since the most recent phase shift prior to this year's event was in the 1970's. The new effects will be noticed mostly by those living in Midwest United States... and the Arctic (which is consequently warmer than usual). I wonder how the polar bears will fare this year with a limited amount of ice formation...&lt;br /&gt;&lt;br /&gt;Let's not think about the diminishing polar bear population and instead discuss something more fun, like biochemistry. In a class I took called "20.320, Analysis of Biomolecular Systems" (no really, I'm serious, we're talking about biochemistry), I had a project in which I had to model a regulatory pathway of my choosing that would ultimately increase or decrease under certain conditions the expression of a protein - in this hypothetical case it was GFP. I chose, rather foolishly I suppose, what I thought at first to be a simple system involving microRNA (miRNA). miRNA is a small strand of RNA (around 21-24 nucleotides long) that thus far has been found to "silence" gene expression by binding onto mRNA, thus disrupting translation. In summary, I hypothesized that miRNA that targeted the gene expressing GFP would be increased by a transcription factor (the transcription factor binding step was not important for the model), but would be regulated downstream during translation by the disruption of miRNA binding to target mRNA, consequently increasing GFP expression, which makes sense - disrupt the disruptor and you get positive activity. It turns out there have been several studies on endogenous regulators of miRNA silencing (called RNA interference in general), most of which involve either the dicing of miRNA itself or the simple removal of miRNA from the cytosol (by specified ribosomes). At this point I saw it would be more efficient if there was some other mechanism that would link miRNA production to GFP DNA production so that so much miRNA and DNA didn't have to be wasted, but I couldn't find any relevant literature. Having no other explanation at the time, I was begrudgingly satisfied and went on with the project, which ended up being a pain versus a simpler model I could've chosen, which was a model of transcription factors similar to the lac operon (but everyone else was doing this, and I didn't want to be everyone else! House. Road House.). The pain was that at the same time as GFP was being produced, miRNA was also disrupting GFP production, which made the system a parametric one of dimension n-squared versus a linear system... and I had to solve the differential equations for all of that shit (about 18 different equations with 18 unknowns in all)!&lt;br /&gt;&lt;br /&gt;Anyways, why I'm talking about this now is that I read in the most recent &lt;i&gt;Cell&lt;/i&gt; issue about a study that could have majorly simplified my model and also my life. This: &lt;a href="http://www.cell.com/fulltext/S0092-8674%2809%2901570-0"&gt;Transcriptional Control of Gene Expression by MicroRNAs&lt;/a&gt;. To summarize, Basel Khraiwesh et al found that miRNA use epigenetic control by methylating DNA transcription sites (making them inactive) to regulate gene expression at high concentration. They also found that at low concentration, the standard RNA/translational control applies, which says this mechanism may be dependent on the miRNA-to-target ratio. The study was done in moss, but the hope is that it translates to mammals. I'm impressed because this also implies that instead of seeing miRNA as a second-son of gene regulation relative to transcription factors, it's just as important, if not more so. Now if only they'd published this a year earlier...&lt;br /&gt;&lt;br /&gt;Sorry guys for that block of science. I'll go back to being foolish.&lt;br /&gt;&lt;br /&gt;I braved the cold to seek liquid warmth in a bar crawl with a few friends in Boston. Before the expedition began, we determined the best way to know if a place is good is by the quality of their french fries, a true American staple. To his credit, one, Don Schmierer, says he is greatly repentant. A law that puts human life at risk without imminent threat with proof of causality is a bad one. In fact &lt;i&gt;she was a fan&lt;/i&gt; and we had a conversation about how to survive a zombie apocalypse. On that note, I need to procure some MREs and a police baton because we all know that &lt;a href="http://www.blogger.com/goog_1262373419706"&gt;police batons &lt;/a&gt;&lt;a href="http://hellforge.gameriot.com/blogs/Hellforge/Left-4-Dead-2-Weapon-Effectiveness-vs-Tank"&gt;do the most damage to a Tank in Left 4 Dead 2&lt;/a&gt;, up there with frying pan. The book delivers. If you need any pointers on how to notice if your household robot is going rebellious, ask me and we can meet within a Faraday cage to speak details.&lt;br /&gt;&lt;br /&gt;Digging through some old tracks, I found the original samples taken for Beyonce's "Crazy in Love" and a couple of Jay-Z's songs, most notably "99 Problems" and "December 4th". I've shortened the content to 3 sections since things I try and things I do in life are pretty much the same&lt;br /&gt;&lt;/span&gt;&lt;/i&gt;&lt;br /&gt;&lt;br /&gt;&lt;b&gt;&lt;span style="font-size: large;"&gt;01 // In&lt;/span&gt;&lt;/b&gt;&lt;br /&gt;&lt;br /&gt;These music videos highlight an interesting cell-shading-type animation style that is similar to, if not modeled after, the Japanese &lt;a href="http://en.wikipedia.org/wiki/Ukiyo-e"&gt;ukiyo-e style&lt;/a&gt;. This makes for a visually intoxicating (fun) and trippy experience. All of these videos are Japanese groups, and I'll explain as you move through. Go ahead and &lt;strike&gt;do &lt;i&gt;whatever it is you do&lt;/i&gt; to&lt;/strike&gt; prep for this, it won't be enough!&lt;br /&gt;&lt;br /&gt;&lt;b&gt;HiFana -&amp;nbsp; Wamono&lt;/b&gt;&lt;br /&gt;&lt;span style="font-size: small;"&gt;HiFana is a beat-mixing duo that features percussion instruments (including a full drum set), MPC, and turntables. They pioneered with Pioneer (the company, heh) the DVJ, or DVD/CD turntable deck, and are associated with a break-heavy, hip-hop style. Their music carries a fast, bouncy beat with some synth and guitar sounds and a layer of rap to balance everything out. Their songs are consistently exposed to Japanese popular culture in the form of anime TV series, videogames and movies, most notably as part of the release of "Kung Fu Hustle" in Japan. This video's more tame/conventional. They also feature a rhythmic lyrical style over a fast beat, but their rock-edge is harder with more guitar riffage and melodic vocals. Atarashii sekai means "new world"; the song is part of an album that revolves around a concept world. There were a lot of cool jazz LPs/EPs as a decent collection of rock/pop. They also had a table full of boxes with really cheap singles or albums they marked down to 3 for $1. $20 got me a Duke Ellington, Van Halen (not what you'd think is my cup of tea, but I like the heavy guitar, yo), a Destiny's Child single (really happy about this one), a Beatles compilation (I don't know what's on it, but I'm hoping it has "I'll Follow the Sun"), and a bunch of dollar records including two Japanese ones, one of which is a single by Ayumi Hamasaki!&lt;br /&gt;&lt;br /&gt;Coincidence: while I was clearing out old stuff from the house, I discovered my dad's old stash of vinyl records, one of which was the other half of The Beatles compilation series (sweet). Besides a bunch of oldies (Neil Diamond, The Beatles, various pop stars of "the ancients"), there were a few that need to be mentioned from the collection.&lt;br /&gt;&lt;ul&gt;&lt;li&gt;Kansas (the original album)&lt;/li&gt;&lt;li&gt;Nat King Cole (&lt;i&gt;Unforgettable&lt;/i&gt;)&lt;/li&gt;&lt;li&gt;a Stevie Wonder album&lt;br /&gt;&lt;/li&gt;&lt;li&gt;A Midsummer Night's Dream (awesome)&lt;/li&gt;&lt;li&gt;A Holiday with&lt;b&gt; &lt;/b&gt;Kenny Rogers (we've got a winner!)&lt;br /&gt;&lt;/li&gt;&lt;/ul&gt;I've got a lot of new acetate now :D. Besides finding about a new word of which I've been missing the opportunity to utilize more regularly for several years, CNN and the Pew focus group found that "teens sext to people whom they are in a relationship, but often times (where the danger lies) [these messages] are forwarded to people outside the relationship, especially after a breakup." If it leaves a paper trail, don't do it, is what my Godfather used to say.&lt;br /&gt;&lt;br /&gt;&lt;a href="http://www.people.com/people/article/0,,20331291,00.html"&gt;Tiger Woods scandal&lt;/a&gt; - I only keep up with celebrity news on the occasions when it involves people I like (i.e. the Rihanna scandal) or just simply blindside me (in essence it'd be or exceed a kiloChi). So Tiger Woods. Besides him crashing his SUV into a tree because he was on Vicadin and Ambien before he drove, women all over have been popping up claiming he had an affair with them. There are allegedly 7-8 women now implicated, according to &lt;i&gt;People&lt;/i&gt;. This gives new meaning to the phrase, "Go get 'em, Tiger." The tragedy is with Elin, his wife, and their son and daughter, although most direct sources to Elin and Tiger say they're trying to get through the mess while preserving the family. It sounds like most of the credible cases with Tiger involve him contacting them frequently, although the ratio of romance to tryst of these meetings is still under scrutiny. A lot of these women surfacing are "party girls" and are probably just jumping onto the bandwagon for the money. The hotel was decked out in Christmas gear and everyone was nice so I couldn’t get too annoyed. Also I won a Premier Line pass in the airport coming back in, although I already had Premier Exec… so I got playing cards instead.
